Movie pays homage to the classic by Sir Arthur Conan Doyle, Sherlock Holmes, with bold interpretations of the iconic characters, Sherlock Holmes and Dr. John Watson. MISS SHERLOCK is set in modern day Tokyo and both lead characters are Japanese women - Dr. Wato Tachibana/Shihori Kanjiya, a surgeon recently returned from a volunteer doctors' mission in Syria and Sara Shelly Futaba/Yuko Takeuchi "Miss Sherlock", an investigation consultant to the police department who solves bizarre and difficult cases.

Throughout the series, the pair solves mystery after mystery with Miss Sherlock's extraordinary observation and reasoning skills.

Akatsuka Detective Agency stands inconspicuously at Shimo Akatsuka in Tokyo’s Itabashi ward. Its carefree, eccentric investigator Nanase Goro is eager to take on troublesome cases that everyone else is reluctant to. At times, he and his partner Kogure Kyusaku tackle sad cases and sometimes difficult ones that are beyond imagination. The detective agency is headed by Kaze Kaoru, a heavy drinker who downs alcohol during the day but watches out for Goro and Gu-re.

Shinichi and Ran's class decides to take a trip on a boat, but Shinichi decides against going. He quickly changes his mind when he finds a letter of challenge stating that one kid will be kidnapped during the trip. He is eager to accept the challenge, but when the one that vanishes is their friend Sonoko, Shinichi must use all of his reasoning abilities to find Sonoko and protect the other students. First he doesn't succeed, he receives a call from the kidnapper stating he will now abduct another student at exactly twelve noon. Shinichi must now recover from his previous failure and hope to prevent a tragedy. Shinichi receives a letter from an unknown challenging him to stop the kidnapping from happening.
